Introduction of VELOCE™ Cell-Free Protein Expression System

Concept

The cell-free protein expression (CFPS) system is an in-vitro transcription/translation platform that uses exogenous DNA or mRNA templates. Proteins are synthesized in clarified cell extracts or fully reconstituted mixtures containing the necessary ribosomes, enzymes, cofactors, amino acids, and energy components—without relying on living cells. Free from cellular metabolism and compartmental constraints, CFPS enables rapid, scalable protein production. Common formats include E. coli lysate, insect cell lysate, wheat germ, and rabbit reticulocyte systems.

This kit supports fast, high-efficiency expression of target proteins from plasmid or linear DNA templates. It employs a coupled transcription–translation workflow to produce protein in a single, “one-pot” reaction. The ready-to-use mix provides all reagents required for in-vitro expression—enzymes, energy system, amino acids, nucleotides, and inorganic salts—offering simple setup and robust yields.

Advantages

  • Speed — Expression in as little as 1 hour (protein/template dependent).
  • Template flexibility — Supports linear DNA, plasmids, and PCR products (no cloning required).
  • High yield & scalability — Achieves >3 mg/mL protein; run in 96-well plates for screening and scale up after hits.
  • Parallelization & co-expression — Miniaturized reactions enable simultaneous expression of multiple proteinsin the same run or the same mix.
  • Simple one-pot workflow — Add DNA to the master mix; no transformation, culture, harvest, lysis, or centrifugation.
  • Toxic or hard targets — No cell-viability constraints; suitable for host-toxic proteins, multi-pass membrane proteins, and disulfide-rich proteins.
  • Open, tunable system — Easily adjust conditions (e.g., salts, cofactors, chaperones) to control transcription/translation/PTMs and reduce inclusion bodies.
  • Built-in labeling options — Straightforward incorporation of non-natural or isotopically labeled amino acids for specialized proteins.
  • Fast turnaround for discovery — Ideal for high-throughput ligand screening and proteomics with rapid iteration cycles.
